Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. It corresponds to the ACTH(4-7) fragment extended at the C-terminus by a Pro-Gly-Pro tripeptide, an addition intended to slow enzymatic breakdown. The molecular formula is C37H51N9O10S and the molecular mass is approximately 814 daltons. In the literature it is often described as an ACTH(4-10) analog, although that label reflects a naming convention as much as a precise structural relationship. The compound was developed in Russia and remains most closely associated with that research tradition.
Laboratory descriptions of the material are consistent across suppliers. It appears as a white to off-white lyophilized powder that dissolves readily in water and in polar organic solvents such as dimethyl sulfoxide. Aqueous solutions are clear and colorless at low concentrations. Because the peptide contains methionine, oxidation at the sulfur atom is a recognized degradation pathway, and handling notes usually call for protected, desiccated storage. Reported purity for research-grade lots is generally above 95 percent as measured by reversed-phase high-performance liquid chromatography.
Russian regulatory authorities approved the peptide for nasal administration, and it remains listed in the national pharmacopoeia under several trade names. Documented indications include acute ischemic stroke, transient ischemic attacks, traumatic brain injury, and certain ophthalmological and neurological conditions. Physicians also prescribe it for cognitive complaints in older patients, although the evidence base for that use is thinner. Outside Russia and a few neighboring states, the substance is not an approved medicine and is sold instead as a research chemical.
Later generations of the molecule include an N-acetylated form and an amidated form, both marketed online alongside the parent peptide. These variants differ in terminal chemistry and stability, and they are frequently discussed in the same breath even though they have not been compared in controlled trials. Supply outside formal healthcare systems comes largely from laboratories that synthesize peptides to order. Purity and identity of these materials vary widely, and no single body oversees the international trade.

Regulatory status varies sharply by country. Semax is registered for medical use in Russia, where it appears in formularies as a nasal solution, and it also holds registration in a small number of neighbouring states. It has no approval from the United States Food and Drug Administration or the European Medicines Agency, and it is not a scheduled controlled substance in most jurisdictions. Elsewhere it circulates mainly as laboratory material, so purity documentation comes from suppliers rather than from a national pharmacopoeia.
Semax is a synthetic peptide created in the Soviet Union during the early 1980s by researchers working in Moscow. It was built from the short adrenocorticotropic hormone fragment known as ACTH(4-10), and the chain was then extended with three additional amino acids. The resulting molecule was named semax and entered clinical use in Russia in 1994. It is generally described as a nootropic and neuroprotective agent rather than as a hormone analogue.
The parent fragment ACTH(4-10) carries the sequence Met-Glu-His-Phe-Arg-Trp-Gly. Semax replaces the arginine and tryptophan positions with a proline-glycine-proline tail, giving Met-Glu-His-Phe-Pro-Gly-Pro. That change removes residues associated with adrenal stimulation, so the peptide does not drive cortisol release the way full ACTH does. This distinction shapes how the compound is grouped in the literature, where it sits with neuropeptides and peptide neuromodulators rather than with corticosteroids.
When a drug is developed with evidence throughout its history of research to show it is safe and effective for the intended use in the United States, the company can file an application – the New Drug Application (NDA) – to have the drug commercialized and available for clinical application. NDA status enables the FDA to examine all submitted data on the drug to reach a decision on whether to approve or not approve the drug candidate based on its safety, specificity of effect, and efficacy of doses.
In contrast with the previous study on PFK1, this study found that elevating OGT or O-GlcNAc upregulated PFK1, though the two studies are consistent in finding that O-GlcNAc levels are positively associated with flux through the pentose phosphate pathway. This study also found that decreasing O-GlcNAc selectively killed cancer cells via ER stress-induced apoptosis. Human pancreatic ductal adenocarcinoma (PDAC) cell lines have higher O-GlcNAc levels than human pancreatic duct epithelial (HPDE) cells. PDAC cells have some dependency upon O-GlcNAc for survival as OGT knockdown selectively inhibited PDAC cell proliferation (OGT knockdown did not significantly affect HPDE cell proliferation), and inhibition of OGT with 5S-GlcNAc showed the same result. Hyper-O-GlcNAcylation in PDAC cells appeared to be anti-apoptotic, inhibiting cleavage and activation of caspase-3 and caspase-9. Numerous sites on the p65 subunit of NF-κB were found to be modified by O-GlcNAc in a dynamic manner; O-GlcNAc at p65 T305 and S319 in turn positively regulate other modifications associated with NF-κB activation such as p300-mediated K310 acetylation and IKK-mediated S536 phosphorylation. These results suggested that NF-κB is constitutively activated by O-GlcNAc in pancreatic cancer. OGT stabilization of EZH2 in various breast cancer cell lines has been found to inhibit expression of tumor suppressor genes.
== Adverse effects == Fluvoxamine's side-effect profile is very similar to other SSRIs. Gastrointestinal side effects are characteristic of those receiving treatment with fluvoxamine. However, compared to escitalopram and sertraline, fluvoxamine's gastrointestinal profile may be less intense, often being limited to nausea. Mosapride has demonstrated efficacy in treating fluvoxamine-induced nausea. It is also advised practice to divide total daily doses of fluvoxamine greater than 100 mg, with the higher fraction being taken in the evening (e.g., 50 mg at the beginning of the waking day and 200 mg at bedtime). In any case, high starting daily doses of fluvoxamine rather than the recommended gradual titration (starting at 50 mg and gradually titrating, up to 300 mg if necessary) may increase the likelihood of nausea. In comparison to other SSRIs, fluvoxamine has the second highest rate of causing discontinuation syndrome, as a result of the low half-life of fluvoxamine.
== Medical uses == Pegulicianine is indicated for fluorescence imaging in adults with breast cancer as an adjunct for the intraoperative detection of cancerous tissue within the resection cavity following removal of the primary specimen during lumpectomy surgery.

==== Acne vulgaris ==== As of 2012, evidence for light therapy and lasers in the treatment of acne vulgaris was not sufficient to recommend them. There is moderate evidence for the efficacy of blue and blue-red light therapies in treating mild acne, but most studies are of low quality. While light therapy appears to provide short-term benefit, there is a lack of long-term outcome data in those with severe acne.
Tim Hortons sells coffee, tea, hot chocolate, and soft drinks. In the mid-1990s the chain moved into specialty and premium items such as flavoured cappuccino, iced cappuccino, and iced coffee. Despite an expansion in their food offerings around 2009, the brand remained heavily dependent on coffee sales. In 2009, it was reported that 60% of their sales occur in the morning, and of that more than 50% is coffee. The coffee served is a blend of 100% arabica beans. The "original blend" is a medium, balanced roast that is the most popular served coffee in Canada. The chain has an "always fresh" policy where coffee is served within 20 minutes of brewing. In November 2011, the company announced that they would be adding espresso machines in 1,000 of their locations, later that month. On April 16, 2012, the brand launched frozen lemonade, in two flavours: original and raspberry. On August 15, 2014, the company added a dark roast coffee blend as an alternative.
==== PEG treatment ==== The purpose of the polyethylene glycol is to remove water from the wood while simultaneously bulking the deteriorated wood cells. PEG is applied to the surface, either by spraying or immersing the object in a solution of PEG in water. Over the course of the treatment, PEG is slowly increased in concentration, pushing out the excess water, coating the interior walls of the wood cells with PEG, which, depending on the molecular weight selected, will provide support once the artifact is dried. PEG compounds with different molecular weights may be used depending on the wood type, level of deterioration, and scope of project. The PEG treatment is often paired with vacuum freeze-drying, as the eutectic point of most PEG solutions is below the freezing point of water. This allows the "free" waterlogging water to sublimate in the process of freeze-drying, which minimizes damage to the wood by preventing ice from forming within the cells and expanding, causing more shrinking and warping. Some artifacts are too large to fit in a commercially available freeze-drier (see the treatment for Vasa or Mary Rose or the Bremen cog).
The Diabetes Research in Children Network Study Group observed blood sugar levels decrease rapidly in the first 15 minutes of exercise and continue to drop during the 75-minute session. The Diabetes Research Group also found that after participating in prolonged aerobic exercise, 83% of participants had at least a 25% decrease in blood sugar levels. High-intensity and concurrent training interventions, strength training, and prolonged aerobic exercise all have been shown to help reduce HbA1c and blood glucose levels in children with type 1 diabetes; therefore, demonstrating that exercise plays a vital role in the management of type 1 diabetes.
The biosynthesis of melatonin in animals involves a sequence of enzymatic reactions starting with L-tryptophan, which can be synthesized through the shikimate pathway from chorismate, found in plants, or obtained from protein catabolism. The initial step in the melatonin biosynthesis pathway is the hydroxylation of L-tryptophan's indole ring by the enzyme tryptophan hydroxylase, resulting in the formation of 5-hydroxytryptophan (5-HTP). Subsequently, 5-HTP undergoes decarboxylation, facilitated by pyridoxal phosphate and the enzyme 5-hydroxytryptophan decarboxylase, yielding serotonin. Serotonin, an essential neurotransmitter, is further converted into N-acetylserotonin by the action of serotonin N-acetyltransferase, using acetyl-CoA. The final step in the pathway involves the methylation of N-acetylserotonin's hydroxyl group by hydroxyindole O-methyltransferase, with S-adenosyl methionine as the methyl donor, to produce melatonin. In bacteria, protists, fungi, and plants, the synthesis of melatonin also involves tryptophan as an intermediate but originates indirectly from the shikimate pathway. The pathway commences with D-erythrose 4-phosphate and phosphoenolpyruvate, and in photosynthetic cells, additionally involves carbon dioxide. While the subsequent biosynthetic reactions share similarities with those in animals, there are slight variations in the enzymes involved in the final stages.

=== Comparison with actions of other PDE5 inhibitors === Sildenafil and vardenafil inhibit PDE6, an enzyme found in the eye, more than tadalafil. Some sildenafil users see a bluish tinge and have a heightened sensitivity to light because of PDE6 inhibition. Sildenafil and vardenafil also inhibit PDE1 more than tadalafil. PDE1 is found in the brain, heart, and vascular smooth muscle. It is thought that the inhibition of PDE1 by sildenafil and vardenafil leads to vasodilation, flushing, and tachycardia. Tadalafil inhibits PDE11 more than sildenafil or vardenafil. PDE11 is expressed in skeletal muscle, the prostate, the liver, the kidney, the pituitary gland, and the testes. The effects on the body of inhibiting PDE11 are not known.
== Evolution == 3-Phosphoglycerate dehydrogenase possesses less than 20% homology to other NAD-dependent oxidoreductases and exhibits significant variance between species. There does appear to be conservation in specific binding domain residues, but there is still some variation in the positively charged active site residues between variants. For example, Type III PHGDH enzymes can be broken down into two subclasses where the key histidine residue is replaced with a lysine residue.
=== Automobile airbags and aircraft evacuation slides === Airbag formulations through late 1990s to early 2000s contained mixtures of oxidizers, sodium azide and other agents including ignitors and accelerants. An electronic controller detonates this mixture during an automobile crash:
==== Elimination ==== Psilocybin is eliminated 80% to 85% in urine and 15 to 20% in bile. It is excreted mainly in urine as psilocin-O-glucuronide. The drug was eliminated approximately 20% and 80% as psilocin O-glucuronide in different studies. The amount excreted as unchanged psilocin in urine is 1.5 to 3.4%. Studies conflict on the deaminated metabolites of psilocin, with one study finding that only 4% of psilocin is metabolized into 4-HIAA, 4-HIAL, and 4-HTOL and another that psilocybin is excreted 33% in urine as 4-HIAA. Findings also conflict on whether psilocybin can be detected in urine, with either no psilocybin excreted or 3% to 10% excreted as unchanged psilocybin. A majority of psilocybin and its metabolites is excreted within three hours with oral administration and elimination is almost complete within 24 hours. The elimination half-life of psilocybin, as psilocin, is 2.1 to 4.7 hours on average (range 1.2–18.6 hours) orally and 1.2 hours (range 1.8–4.5 hours) intravenously. Psilocin's elimination half-life in mice is 0.9 hours, much faster than in humans. Psilocin O-glucuronide's half-life is about 4 hours in humans and one hour in mice. No dose adjustment of psilocin is thought to be required as psilocin is inactivated mainly via metabolism as opposed to renal elimination. Accordingly, glomerular filtration rate (GFR) did not affect the pharmacokinetics of psilocybin.
=== Casting === In February 2025, Jessica Lange, who starred in the first four seasons and briefly appeared in the eight season in 2018, answered a question about her potential return for the thirteenth season: "Oh Christ, no. I mean, I haven't done it for more than 10 years, 12 years, so, no, I'm not doing it." Murphy revealed that Lange was the first person he turned to about the return and she agreed under the condition she would have a musical number. Subsequently, he turned to Sarah Paulson and Evan Peters, who agreed instantly. They both last appeared in the tenth season in 2021. In October 2025, Lange, Paulson, Peters, Emma Roberts, Billie Lourd, Kathy Bates, Angela Bassett, Leslie Grossman, Jessica Lange, and Gabourey Sidibe were all confirmed to return for the thirteenth season, along with the newcomer Ariana Grande. Bates, Bassett and Sibide last appeared in the eight season in 2018. Bates revealed that she would have only one day of filming. In February 2026, John Waters joined the cast, while Joey Pollari joined in April, and Alex Consani and Paul Anthony Kelly joined in May. John Carroll Lynch and Matt Fraser were also confirmed to reprise their roles from Freak Show, with Mena Suvari and Berto Colón also joining the cast. In July, Frances Conroy was confirmed to reprise her role from Coven, with Avantika, Grace Dumdaw, Cara Delevingne, and Charlie Carver also joining the cast. In July 2026, it was reported that Grande dropped out due to scheduling conflicts with the season's production schedule and her Eternal Sunshine Tour.

It is registered as a pharmaceutical product in Russia, where it is typically supplied as a nasal solution. In most other countries it is not an approved drug and is traded as a research chemical instead. Approval status depends entirely on the jurisdiction involved.
Most published work uses intranasal administration, matching the route of the registered Russian formulation. Some animal experiments use intravenous or intraperitoneal injection. The route matters because the intact peptide is cleared rapidly from blood, so exposure differs between methods.
A purity value describes the share of the powder accounted for by the target peptide, usually by high-performance liquid chromatography. It does not confirm that the sequence is correct, which requires mass spectrometry or amino acid analysis. Purity and identity are separate checks.
It originated at the Institute of Molecular Genetics in Moscow during the 1980s. The work was carried out by a Russian research group that specialized in peptide neuropharmacology.
